1. Users provide following information at the beginning of the experiment: Name, PI, Department, Tel., Contact Email, and Recharge Number, and the Number of Experiments (cartridges) to be used.
2. No Radioactive, Acutely Toxic, Toxic, Corrosive, or Flammable materials are allowed in the Instrument
3. Users are responsible for purchasing their own supplies, buffers biosensors and assay plates as listed on the Cortopassi lab website: https://www.cortopassilab.com/octet-instrument.html
4. Users prepare all necessary reagents, pipet their experimental components onto their assay plates, and bring these to the facility. If User needs to prepare the assay plates and cartridges in the Cortopassi lab space we can make arrangements to facilitate Users upon User request that should be made in advance.
5. Instrument Operator login to the Instrument Controller Software and make sure the User can execute the experiment. Instrument Operator can help Users to load the cartridges into the Instrument.
6. Users are required to be familiar with UC Davis Safety policies and training, adhere to UC Davis Safety rules, and be able to provide appropriate documentation. Users are required to have appropriate PPE while in the laboratory space; the facility have extra lab-coats and gloves to facilitate Users.
7. Users will be charged according to the Number of Experiments performed.
Seahorse Assistance, Hourly Labor Rates
The Seahorse XF flux analyzer equipment is available for recharge use at the UC Davis. The 24-well Seahorse system provides researchers the opportunity to perform medium-throughput, real-time measurement of mitochondrial function and cellular bioenergetics in an expedited manner compared to traditional techniques. Seahorse XF Analyzers measure oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) of live cells in a multi-well plate, interrogating key cellular functions such as mitochondrial respiration and glycolysis. XF Analyzers perform compound addition and mixing, label-free analytical detection, and automatic measurement of OCR and ECAR in real time (https://www.agilent.com/en/products/cell-analysis/seahorse-analyzers).

Seahorse XF-24 Analyzers measure OCR and ECAR of live cells in a 24-well plate format, Seahorse-XFe96 – in 96-well format. OCR and ECAR rates are key indicators of mitochondrial respiration and glycolysis and these measurements provide a systems-level view of cellular metabolic function in cultured cells. The description of the systems can be found: https://www.agilent.com/en/product/cell-analysis/real-time-cell-metabolic-analysis/xf-analyzers
